分子对接
从100,000杂交瘤细胞中提取总RNA,RT-PCR扩增出单抗的轻重链核苷酸序列,IMGT/V-QUEST分析抗体序列。
Molecular Operating Environment (MOE) v2018.01软件用于抗体分子的模建。ClusPro用于进行抗原-抗体分子的对接。
A:MAb 3F11和蛋白HAdV11FK之间的结合界面. 分子间有强烈相互作用的位点用stick小棍图来显示,抗体的位点标记绿色,抗原位点标记兰青色。有很强的极性相互作用,如氢键或者盐桥,用浅蓝色破折线标记。图中可见HAdV11FK蛋白的R249,K251,E252和单抗3F11的CDR H1/H2区有很强的极性作用。
